1039 Search for potential disease drivers of hidradenitis suppurativa in non-lesional skin by whole transcriptomic analysis

Hidradenitis suppurativa (HS) is a chronic, inflammatory skin disease of the apocrine gland-rich (AGR) areas. Initial molecular events in HS are poorly explored. We aimed to identify potential driver factors during development by comparing 9 non-lesional (HSNL) 12 region-matched controls RNASeq. 611 significantly differentially regulated DEGs were identified, which 212 upregulated while 399 downregulated. In order gene sets with different expression patterns, KMeans clustering was carried out Morpheus, allowed us distinguish 6 modules. Two contained downregulated genes 4 consisted genes. To understand functional characteristics each module, we performed enrichment analysis on their respective String. Downregulated modules related cellular metabolism and extracellular matrix organization. Among modules, first associated keratinocyte differentiation antimicrobial peptides. The second B cell function, third connected IL-17 pathway chemokine signaling, no found last module. According literature data our own results, two transcription (TFs) (STAT1 GRHL3) have significant role progression HS, therefore, curious if these TFs represented among between HSNL control. Therefore, TF ChEA3 that almost 10% (58 611) GRHL3, 51 only 7 14% (86 STAT1. Both important keratinocytes (KCs), STAT1 regulates immune responses microbes, GRHL3 promotes KC differentiation. These findings further strengthen KCs’ primary initiation indicate as new drug targets.
